As a novel immunotherapy, APVO436 is designed to engage the immune system to mount a targeted response against CD123-expressing hematological tumors. Cytokine release syndrome (CRS) is a significant concern with T-cell activating therapies and has been associated with severe complications in clinical trials. In preclinical studies, APVO436 induced lower levels of several key T-cell cytokines, including IFNg, IL-2, IL-6, and TNFa. Aptevo believes that the improved cytokine activation profile observed in preclinical studies of APVO436 suggest that it could offer a potential safety advantage with reduced toxicities compared to other CD123 x CD3 T-cell engagers at comparable or higher doses.

Preclinical data show that APVO436 is a potent inducer of redirected T-cell killing of AML tumor cells both in vitro and in vivo. When compared head-to-head against an Aptevo-generated version of a competitor molecule, (Macrogenics’ CD123 x CD3 dual-affinity re-targeting (DART) molecule, MGD006), the data show that both were effective at stimulating a tumor-directed immune response and induced comparable T-cell activation, proliferation and cytotoxicity. However, APVO436 induced lower levels of several key T-cell cytokines, including IFNg, IL-2, IL-6, TNFa, which have been associated with serious adverse events in clinical studies.
The APVO436 Phase 1/1b clinical trial will be conducted in two parts. The first part will enroll up to 60 patients and is an open-label, dose-escalation study evaluating the safety and pharmacokinetic profile of APVO436 to determine a recommended dose for part 2. The second part of the study is a Phase 1b open-label expansion study to assess the clinical activity and safety profile of APVO436 at the recommended dose in a larger group of patients.

Pope Francis tells Catholic TV journalists to be 'channels' of peace
Vatican City, Dec 13, 2018 / 09:55 am (CNA/EWTN News).- Pope Francis told a Catholic media group Thursday to be avenues of God’s peace, sharing the stories of the poor, the least, and the voiceless.
“In your profession you can be ‘living channels’ of spirituality before God and before all your listeners and viewers,” the pope told collaborators of the Italy-based Catholic broadcasting network Telepace.
“I renew, then, the invitation to ‘promote a journalism of peace,”” he said, quoting his 2018 message for World Communications Day. “A journalism created by people for people, one that is at the service of all, especially those – and they the majority in our world – who have no voice.”
Francis addressed the group Dec. 13, for their 40th anniversary, in the Vatican's Clementine Hall. Since 1990, at the request of St. John Paul II, the network has broadcast Vatican events such as the general audience, the Angelus, and papal Masses.
Praising the network, the pope said he wanted to urge three commitments in journalism – the first, to be “antennas of spirituality.” The TV antenna has a beautiful symbolism, he said, because of its “dual function of emitting and receiving a signal.”
Broadcast journalism should be a voice for the voiceless, he stressed; above all for the poor, the least, and the excluded. “Never forget them, the poor next door!” he said, praising the network’s program about inmates on death row in Texas. This “is the spirituality of charity!” he said.
Urging journalists to consider how they can teach the Gospel to the young, he stated that he would like the media to “pay more attention to young people, not only by telling their failures but also their dreams and their hopes!”
Doing this, he said, is a matter of being witnesses of God’s Word.
He also warned the media against letting their storytelling ever devolve into gossip, which undermines human community and sows “envy, jealousy, and lust for power.”

A quirky French SUV you’ve probably never heard of has a dashboard strikingly similar to the one in Tesla's Model 3 — take a look (TSLA)
· The Tesla Model 3 is famous for its minimal interior and single, central touchscreen.· Other automakers are moving the same direction as Tesla with their designs.
· They aren't common in the US, but I found a very Model 3-esque setup in a Citroën C4 Cactus in Rome.
· The Citroën C4 Cactus is a quirky French compact crossover SUV. The reaction that a lot of people have when they first slip into a Tesla Model 3 is one of mild shock that there isn't an instrument panel, nor much in the way of controls — just a large, central touchscreen that pretty much runs everything.
Revolutionary? Sort of.
Minimizing visual instrumentation and in-car controls isn't new, but Tesla has taken it to an extreme.
Other automakers have also taken the plunge. I was reminded of this recently when I visited Rome, rented a car, and wound up at a Citroën dealership.
